This study investigates the resistance status of the house mosquito, Culex pipiens, in Lebanon, to organophosphate and carbamate insecticides. For this, mosquitoes were collected from different regions in Lebanon from December 2008 to December 2009. Organophosphate resistance in mosquitoes could be caused by two factors: elevated levels of A and B esterases, which detoxify the insecticides, and mutations in the acetylcholinesterase gene, which is the target of the organophosphates and thus the mutation produces a form of insensitivity to these insecticides. A total sample of 345 mosquitoes was collected from the six Lebanese provinces and screened for overproduction of detoxifying esterases. In parallel, 290 mosquitoes were analyzed for G119S and F290v mutations in the ace-1 gene. Our results show that resistance in the Lebanese C. pipiens populations is caused by an overproduction of the A2-B2 esterases and by the mutation G119S. It was found that 38.6% of the sampled mosquitoes exhibited an overproduction of the A2-B2 esterases, whereas 61.4% were susceptible. Moreover, 24.1% of the mosquitoes enclosed the G119S mutation and these were of the heterozygote resistant genotype (RS), while 75.9% were susceptible. The RR genotype, homozygote resistant, was not detected in the sampled mosquitoes because of a high fitness cost associated with the mutated AChE1, enzyme coded by the ace-1 gene. No F290V mutations were found in the Lebanese C. pipiens populations.
