In 1987 the Electron Microscopy Society of America (EMSA) going to drive important scientific discoveries across wide areas under the leadership of J. P. Revel (Cal Tech) initiated a major of physiology, cellular biology and neurobiology. They had been program to present a discussion of recent advances in light looking for a forum in which they could advance the state of microscopy as part of the annual meeting. The result was three the art of confocal microscopy, alert manufacturers to the lim special LM sessions at the Milwaukee meeting in August 1988: itations of current instruments, and catalyze progress toward The LM Forum, organized by me, and Symposia on Confocal new directions in confocal instrument development. LM, organized by G. Schatten (Madison), and on Integrated These goals were so close to those of the EMSA project that Acoustic/LM/EM organized by C. Rieder (Albany). In addition, the two groups decided to join forces with EMSA to provide there was an optical micro-analysis session emphasizing Raman the organization and the venue for a Confocal Workshop and techniques, organized by the Microbeam Analysis Society, for NSF to provide the financial support for the speakers expenses a total of 40 invited and 30 contributed papers on optical tech and for the publication of extended abstracts.

1.1 Overview The precise knowledge of the three-dimensional (3-D) assembly of biological structures is still in its origin. As an example, a widely accepted concept and common belief of the structure of the airway network oflung is that of a regular, dichotomous branching pattern, also known as the trumpet model. This model, first introduced by Weibel in 1963, is often used in clinical and physiological applications. However, if this concept of dichotomy is used to model lung, a shape is obtained that is quite different from a real lung. As a matter of fact, many previous quantitative morphological and stereological investigations of lung did not concentrate on the spatial aspect of lung morphology but delivered data in a more statistical fashion. Accordingly, the functional behavior predicted by such a model becomes questionable and indeed, the morphometrically predicted lung capacity exceeds the physiological required capacity by a factor of 1.3 up to a factor of2. This problem has also been termed a paradox, as discussed by Weibel in 1983. In the rare cases where descriptive models of the mammalian bronchial tree exist, monopodial in small mammals, dichotomous in larger ones, the understanding of the historical and/or functional reasons for size-related changes in the general design is not explainable. This investigation is trying to overcome this gap by computer modeling and functional simulation.

Written by an international team of experts from the Tara Oceans Marine Biology Imaging Platform (TAOMI), this is the first and only compendium on marine imaging technologies, and includes all known underwater as well as on-land techniques. TAOMI is imaging the largest collection of marine organisms in recent history, ranging from viruses to corals, and is duplicated on land to perform high throughput confocal analysis of plankton, X-ray tomography as well as cryo-electron microscopy. This unique platform combines underwater imaging with cytometry, stereomicroscopy, fluorescence microscopy and 3D microscopy - all of which are covered in this practical book, along with remote sensing, MRI, and optical projection tomography. The definitive resource for every marine biologist who is planning to image marine species, whether underwater or on land.

The Handbook of Biomedical Nonlinear Optical Microscopy provides comprehensive treatment of the theories, techniques, and biomedical applications of nonlinear optics and microscopy for cell biologists, life scientists, biomedical engineers, and clinicians. The chapters are separated into basic and advanced sections, and provide both textual and graphical illustrations of all key concepts. The more basic sections are aimed at life scientists without advanced training in physics and mathematics, and tutorials are provided for the more challenging sections. The first part of the Handbook introduces the historical context of nonlinear microscopy. The second part presents the nonlinear optical theory of two- and multiphoton excited fluorescence (TPE, MPE) spectroscopy, second and third harmonic generation (SHG, THG) spectroscopy, and coherent anti-Stokes Raman spectroscopy (CARS). The third part introduces modern microscopic and spectroscopic instrumentation and techniques that are based on nonlinear optics. The fourth part provides key applications of nonlinear microscopy to the biomedical area: neurobiology, immunology, tumor biology, developmental biology, dermatology, and cellular metabolism. There are also chapters on nonlinear molecular probes, cellular damage, and nanoprocessing.

First published in 2006, this book has become the standard reference on nano-optics. Now in its second edition, the text has been thoroughly updated to take into account new developments and research directions. While the overall structure and pedagogical style of the book remain unchanged, all existing chapters have been expanded and a new chapter has been added. Adopting a broad perspective, the authors provide a detailed overview of the theoretical and experimental concepts that are needed to understand and work in nano-optics, across subfields ranging from quantum optics to biophysics. New topics of discussion include: optical antennas; new imaging techniques; Fano interference and strong coupling; reciprocity; metamaterials; and cavity optomechanics. With numerous end-of-chapter problem sets and illustrative material to expand on ideas discussed in the main text, this is an ideal textbook for graduate students entering the field. It is also a valuable reference for researchers and course teachers.
